DETECTION OF COLOMBIAN DATURA POTYVIRUS AND TWO UNIDENTIFIED POTYVIRUSES IN BRUGMANSIA HYBRIDS

Abstract:
Four potyvirus isolates from Brugmansia, and one each from Juanulloa, Lycopersicum and Petunia are assigned to the same virus on the basis of particle morphology, cytopathology and immunoelectron microscopy. The potyvirus isolates reach identical, high decoration titers with original antiserum to Colombian datura potyvirus (CDV) and their heterologous titers with antisera to other serologically related potyviruses (bean common mosaic, carnation vein mottle, endive necrotic mosaic, plum pox, and sweet potato latent potyviruses) differ from the respective homologous titers by five or more dilution steps. Therefore, the new isolates are considered to be CDV although the original isolate is lost. Classification of CDV as definitive member of the genus Potyvirus is confirmed by the serological relationship of the new isolates to several potyviruses. In addition to CDV, two potyviruses are found in several Brugmansia cvs. which do not react with antiserum to CDV and differ from CDV and each other in serological reactions with antisera to potyviruses and in cytopathology.
