CELLULOSE BINDING PROTEIN, A NOVEL TOOL IN BIOTECHNOLOGY DIAGNOSTICS.

Abstract:
Shoseyov and Doi (1990) isolated a unique cellulose binding protein (CBPA) from the cellulolytic bacterium Clostridium cellulovorans. It was found that this major subunit of the cellulase complex has no hydrolytic activity and is essential for the degradation of crystalline cellulose. This protein has an extremely strong affinity to cellulose. More recently we cloned and sequenced cbpA (Shoseyov et al., 1992). Using PCR primers that flank the cellulose binding domain (cbd), we successfully PCR amplified and cloned cbd into an overexpression vector that enables us to overproduce the 17 kDa CBD in E. coli (Goldstien et al., 1993). The recombinant CBD has a strong affinity to cellulose and chitin.

The cellulose binding domain is useful in developing unique products based on bio-immobilization of biologically active molecules to cellulose and chitin. Applications of CBD technology include: CBD-fusion vectors for production and isolation of different recombinant CBD-fusion proteins such as enzymes, hormones, growth factors vaccines and antibodies. The extremely low non-specific protein binding to cellulose matrices, together with the novel CBD-Technology enable us to develop sensitive diagnostic tools.
