A GENERAL TOSPOVIRUS ASSAY USING MONOCLONAL ANTIBODIES AGAINST TOMATO SPOTTED WILT VIRUS GLYCOPROTEINS

Abstract:
The efficiency of the glycoprotein (GP) specific serological assay was compared with nucleocapsid (N) protein specific serological assay by testing samples from 26 ornamental and vegetable species for the presence of tomato spotted wilt virus (TSWV) and impatiens necrotic spot virus (INSV). Of the total of 142 N protein-positive samples, 94% (TSWV) and 97% (INSV) were positive in the GP assay. No N protein-negative samples were GP-positive, confirming the reliability of the new assay. Flowers were a very good source of tissue for detecting tospovirus infections, and even symptomless petals from infected plants gave positive results with N protein-and GP-specific assays. This may help to explain the high vector efficiency of Frankliniella occidentalis, which preferentially feeds on flowers.
