SENESCENCE IN IRIS; IDENTIFICATION OF MOLECULAR MARKERS

Abstract:
The first visible symptom of senescence in Iris flowers (Iris x hollandica Tub., cv Blue Magic) is tepal inrolling, starting on day 4 after harvest. Treatment of flag tepals with cycloheximide delayed the onset of this symptom. Cycloheximide also prevented the decrease in protein content, indicating that de novo synthesis of proteases is a key event in tepal senescence. In an attempt to identify processes involved in tepal senescence several differentially expressed genes have been detected using Differential Display Reverse Transcriptase PCR (DDRT-PCR). These genes will be cloned and analyzed. The DDRT-PCR method will allow identification of minor transcripts, which may be important regulatory factors.
