Abstract:
Somatic embryogenesis has been achieved in 17 out of 24 varieties of roses which we have investigated.
Embryogenic, friable callus was obtained by culturing in vitro roots on SH medium (Schenk and Hildebrandt, 1972) supplemented with 3mg/l 2,4-D, or on half-strength MS medium (Murashige and Skoog, 1962) supplemented with 3mg/l 2,4-D and 600mg/l L-proline.
Direct embryogenesis was obtained from root cultures of three of these cultivars by transfer of the roots to the latter medium.
A combination of cold treatment at 4°C for 2 weeks, the substitution of sucrose by maltose and the use of thidiazuron (0.1 mg/l) as the only growth regulator, stimulated a germination rate of 46% of somatic embryos of Rosa 'Frensham'.
