THE SELF-INCOMPATIBILITY GENE IN APPLE AND DETERMINATION OF THE S-GENOTYPE OF APPLE CULTIVARS BY PCR

Abstract:
Pollen-pistil interactions in apple (Malus X domestica Borkh.) are controlled by a gametophytic self-incompatibility (SI) system that prevents self-pollen from reaching the ovary for fertilization. Several alleles of the highly polymorphic SI gene (S-gene) in apple have been cloned and characterized. Comparison of the derived amino acid sequences of S-alleles in apple reveals that the protein structure is composed of alternating conserved and variable sequence regions. Allele-specific PCR primers, designed to anneal within the variable sequence domains, were used to analyse the presence of the corresponding S-alleles in genomic DNA from various cultivars. Here we report on recent data on the S-genotypes of old and current apple cultivars, determined by PCR typing of the S-alleles.
