EFFECT OF EXPLANT TYPE AND CULTURE MEDIUM ON MICROPROPAGATION OF THREE PISTACIA SPECIES

Abstract:
Explants consisting of shoot tips and nodal segments of three species of pistachio, Pistacia vera, P. mutica and P. terebinthus, were cultured on different medium formulations and a range of concentrations of the cytokinin BAP.

BAP was found to be essential for growth from shoot tips and nodal explants. Although P. mutica gave the most rapid growth, both shoot tips and nodes of P. vera develop the greatest number of leaves and therefore more axillary buds on the shoots produced.

The optimum medium appeared to be MS (1962) supplemented with 5mg/L BAP. For the formation of roots on the new shoot 5 mg/L of the auxin IBA was necessary.
