Abstract:
Globular pro-embryos and somatic embryos of different stages (heart-shaped, cotyledonary) differentiated in pistachio (P. vera). Optimal results were obtained with cotyledon explants cultured on medium containing 10 mg/l 2,4-D when the induction medium was renewed once after 4 weeks of culture, and the calli transferred on the development medium 3 weeks after the renewal.
Two types of calli were identified: only the nodular callus was embryogenic in comparison with the granular callus.
Mucilaginous callus has proven to have better embryogenic quality than the compact callus and permitted an easy establishment of cell suspension.
Secondary embryogenesis and precociously germinating embryos of different morphological types were also observed in pistachio.
