Abstract:
In vivo spectrophotometric assays were conducted on bulb sections of Lilium longiflorum, hybrid 2–9, L. x elegans, hybrid 92–3 and L. speciosum, 'Feeling'. The levels of phytochrome were measured using a dual wavelength spectrophotometer and expressed as the absorbance difference between 730 nm and 800 nm.
The amount of phytochrome (Ptot) was highest in L. longiflorum followed by L. x elegans and L. speciosum. The level of Ptot was highest in the basal plate and phytochrome was not detected in the distal part of the daughter scales.
Dark reversion of far -red absorbing form of phytochrome (Pfr) to the red absorbing form (Pr), and dark destruction of Pfr, were observed in all three Lilium species.
The kinetics of destruction revealed that the half life of Pfr is longer than 24 hours, suggesting that phytochrome in Lilium is mostly type II.
