METHOD, MEDIUM AND INCUBATION TIME SUITABLE FOR IN VITRO GERMINATION OF EGGPLANT (SOLANUM MELONGENA L.) POLLEN

Abstract:
Suitable method, medium and incubation time for in vitro pollen germination of eggplant were investigated. "Agar in petri" method gave better result than "hanging drop" and "saturated petri" methods. The most convenient germination medium was 1% agar, 12% sucrose, 300 ppm H3BO3 and 300ppm Ca (NO3)2. Although percent germination was high in this medium, busrting of pollen grains and tubes was very frequent. Several incubation times were studied and 2–3 hours at 25°C was found to be the most suitable time for counting the germinated pollen grains with no bursting. Suitability of the method, the medium, and the incubation time for evaluating pollen fertility was confirmed by comparing pollen germination results with pollen viability estimated by TTC-staining test in 11 eggplant F1 hybrids.
