Abstract:
East Malling is investigating isoenzyme markers in cherry, especially to construct genetic linkage maps for use in its breeding programmes.
Two progenies, Prunus avium 'Napoleon' x P. incisa and 'Napoleon' x P. nipponica, were analysed using polyacrylamide gel electrophoresis for 30 enzyme systems.
Forty-three loci were detected on the basis of segregating zymograms, and 14 putative loci on the basis of polymorphisms.
Analysis of cosegregation revealed that 29 loci can be grouped into 7 linkage groups.
Recombination fractions were calculated and a linkage map is presented.
Backcross progenies have been raised which will segregate for most of the putative loci and may provide additional linkages.
Analysis of 14 cultivars of P. avium for the same 30 enzyme systems showed just 18 of the loci to be polymorphic and revealed two additional putative loci; this indicates far less scope for linkage analysis in intraspecific P. avium progenies.
