PRELIMINARY STUDY OF TULIP PROTEINS DURING SENESCENCE

Abstract:
Conditions for obtaining soluble proteins from petals were defined in order to extract the bulk of soluble proteins without exposing them to denaturation. Electrophoresis, including several enzymatic characterizations performed after electrophoretic separation, were adapted to this study.

The protein content of extracts of petal tissue, which increases until full bloom of the flower, decreases as senescence develops. Qualitative and semi-quantitative data determined from the electrophoretic patterns for glutamate dehydrogenase, glucose-6-phosphate dehydrogenase, malate dehydrogenase, malic enzyme, acid phosphatase, esterase and peroxidase indicate the polymorphic nature of most of these enzymes and show clear cut variation during senescence. The activiites of glutamate dehydrogenase, glucose-6-phosphate dehydrogenase, malic enzyme, peroxidase, which are very discrete or undetectable in extracts of pre-senescent petals, become very marked as senescence develops.
