FACTORS INFLUENCING ROOTING OF PEACH SHOOTS CULTURED IN VITRO

Abstract:
This study describes factors influencing In Vitro rooting of Nemaguard and Meet-Ghamre Peaches using Murashige and Skoog (MS) medium (half strength). The highest rooting percentages were obtained in the cultures media supplemented with either NAA at 2.0 mg/L, IAA at 1.5 mg/L and IBA at 1. or 1.5 mg/L as compared with other concentrations. Increasing the concentration of auxins from 0.5 to 2.5 mg/L positively affected the average number of roots/explant. Rooting percentage was improved by adding 3 g/L activated charcoal (AC) to rooting media supplemented with high concentrations of IAA, NAA and IBA. Dark incubation at 5°C for 2 weeks followed by incubating in 16-h photoperiod at 25°±2°C enhanced the rooting ability of shoots.
