BROCCOLI AND CAULIFLOWER CULTIVARS IDENTIFICATION THROUGH IE-HPLC SEED PROTEIN ANALYSIS

Abstract:
Ion-exchange high performance liquid chromatography (IE-HPLC) for the identification of broccoli (Brassica oleracea L. var. italica Plenck) and cauliflower (Brassica oleracea L. var. botrytis L.) cultivars was investigated, Water-and salt-soluble proteins extracted from unimbibed seeds of 44 cultivars were eluted from an anion-exchange DEAE-825 column with an increasing ionic strength. Proteins were monitored at 254 and 280 nm. For some cultivars the effect of different growing locations on seed proteins was also considered. Qualitative and quantitative analysis of IE-HPLC seed protein profiles obtained from analyzed B. oleracea L. cultivars permitted a clear differentiation among them. Location of seed production had no effect on the elution profile. IE-HPLC seed protein analysis proved to be a high resolution method for broccoli and cauliflower varietal identification and genetic purity assessment. The method is cost effective and time saving.
