Abstract:
In reder to apply gene transfer techniques to chrysanthemum (Dendranthema grandiflora Tzvelev.) improvement, silver nitrate tas been used to develop an efficient and direct shoot regeneration system.
Shoot regeneration directly from internode segments of in vitro grown plantlets of cv.
Wu Huan cultured 4 weeks on MS medium supplemented with optimum concentrations of BAP (0.5–2.0mg/1) and NAA (0.2mg/1) was observed at low frequences (<50%). Incorporating 10 and 20 uM silver nitrate into medium with 0.5mg/1 BAP+0.2mg/1 NAA and medium with 1.0–2.0mg/1 BAP+0.2mg/1 Naa. respectively, highly increased shoot regeneration frequency up to 71–82%. but had no significant effect on the number of shoots per internode segment.
In addition to promoting shoot regeneration directly from cells at the cut edges, silver niti\rate also markedly inhibited the development of callus at the cut surfaces.
The mode of action of silver nitrate is discussed.
