Abstract:
Different treatments of chemical mutagen concentration, time and method were tested to study effect of different mutagens on shoots and leaves of apple and cherry.
The results showed that (1) mutant with 50% living rate were obtained by macerating leaf pieces in vitro in 0.2% EMS solution for 1 hour. (2) cherry shoots were macerated in the solution of 0.2% EMS for 1 hour and other in the solution of 0.2% colchicine for 24 hours, apple shoot were 12 hours in the solution of colchicine, and then they were grafted on the rootstock in greenhouse, mutants with 50% living rate, and 2 cherry and 2 apple mutants with forked leaf and opposite leaf were obtained from VO. (3) Jonagored apple leaf pieces in vitro were cultured on the medium with 0.2% EMS for 3 hours and 2 regenerative shoots were obtained.
