IONIC COMPOSITION AND STRENGTH OF CULTURE MEDIUM AFFECT PHOTOAUTOTROPHIC GROWTH, TRANSPIRATION AND NET PHOTO-SYNTHETIC RATES OF STRAWBERRY PLANTLETS IN VITRO

Abstract:
The growth, net photosynthetic and transpiration rates of strawberry (Fragaria × ananassa Duch. cv. Houkouwase) plantlets under photoautotrophic conditions as affected by ionic composition (modified Hoagland & Arnon, MHA, vs. Murashige & Skoog, MS) and strength of the medium were investigated. Three shoots at a rooting stage, each planted in a 0.13×10-4 m3 liquid medium contained in a separate cup, were cultured in 7.5×10-4 m3 polycarbonate vessels. The vessel had four 10 mm diameter gas permeable microporous filters (number of air exchanges: 3.5·h-1). Cultures were maintained in a room for 21 days at 24°C, 70% RH, 1.0 mmol-mol-1 CO2, 16 h-d-1 photoperiod and 160 μmol ·m-2·s-1 photosynthetic photon flux density provided by cool-white fluorescent lamps. Fresh and dry weights per plantlet on day 21 were greater in the full strength than in the ¼ or ½ strength MHA and MS treatment. Leaf area, shoot to root dry weight ratio, and net photosynthetic and transpiration rates increased with increasing ionic strength in MHA and MS treatments. The medium pH on day 21 was significantly lower in MS (pH 4.4–4.5) than in MHA (pH 6.7–6.8).
