ENHANCEMENT OF SOMATIC EMBRYOGENESIS OF IPOMOEA BATATAS FOR APPLICATION TO A BIOREACTOR PRODUCTION SYSTEM

Abstract:
The physiological age of embryogenic callus influenced the number and overall morphology of the embryo population. Within each age category fewer embryos were formed at 25 than 30 C on semisolid medium. Maximum mature embryo production (35 embryos 10 mg-1 inoculum) was obtained from 6-week-old callus at 30 C. The youngest callus (4 weeks) produced the most immature embryos. Ten- and 12-week-old callus produced fewer immature, fewer mature and more incomplete embryos. Somatic embryogenesis occurred in liquid cultures containing 20 mM NH4NO3 and 30 mM KCl. Globular embryos formed and continued development in suspension producing viable, mature cotyledonary embryos by the 14th day. Initially the gas composition within the Erlenmeyer headspace was that of room air, 0.01 ppm C2H4, 0.03% CO2 and 20.7% O2. Ethylene increased from 0.45 ppm minimum (day 1) to a maximum of 6.37 ppm (day 16). Maximum CO2, 21.2%, was also evident on day 16. Oxygen was depleted from 18.7% (day 1) to a minimum of 8.1% by the 13th day. Mature embryos were evident by day 14 when the O2 had been depleted to less than 10%, and CO2 and ethylene had increased to more than 15% and 5 ppm, respectively.
