MICROPROPAGATION OF GERBERA IN THE “CULTURE PACK”-ROCKWOOL SYSTEM WITH SUGAR-FREE MEDIUM UNDER NON-STERILE CONDITION

Abstract:
Shoots with 2 leaves of Gerbera were planted on Grodan® Rockwool Multiblock (4 by 4 blocks) with sugar-free medium under non-sterile condition in the “Culture Pack” (7.5× 7.5×10.5 cm), a novel disposable box-shaped culture vessel made of fluorocarbon polymer film (Neoflon® PFA films, Daikin Industries, Japan). These culture systems were placed under CO2 enriched condition (3000 ppm). Microbial contamination was not observed on the plantlets and Multiblock in the Culture Pack even when they were planted under non-sterile condition. The plantlets obtained in the Culture Pack-Rockwool system were normal, more vigorous and significantly larger when compared to those on sugar-contained agar medium in bottle, a conventional system of micropropagation. A comparison of the leaf and root morphology of in vitro-cultured plantlets obtained in the both culture systems were also made. There was no problem in using the larger Culture Pack (15×22.5×10.5 cm)-Rockwool sugar-free system to which shoots were planted under non-sterile condition.
