EFFECTS OF MEDIUM PH ON SHOOT GROWTH AND FLOWERING OF TORENIA INTERNODAL STEM SEGMENTS IN VITRO

Abstract:
The effects of medium pH was observed on the shoot growth and flowering of Torenia fournieri Lind. Internodal stem segments were cultured on a defined medium (initial medium pH 5.6) with or without a organic buffer, 2-(N-morpholino) ethanesulfonic acid (MES). There was a obvious negative correlation between the shoot growth and final pH of culture medium after culture for 13 weeks. Then, using unbuffered media, pH of media, top fresh weight and flower bud numbers of each culture were recorded weekly. The medium pH fluctuated apparently, that is, it rose to about pH 7.0 after 5 weeks in culture, and then, it finally dropped to 4.3.

As the results of renewal of medium at different time of culture, it appeared that the first 2 weeks of culture was important period for in vitro flowering of Torenia internodal stem segments.
