A 'SHUTTLE CALLUS SYSTEM'- APPLICATION OF TISSUE CULTURE IN CITRUS

Abstract:
Potential applications of tissue culture to conduct basic research and applied studies in citrus are presented. A 'shuttle callus system', in which embryogenic callus obtained from embryos or embryoids can produce embryoids, and the embryoids in turn can produce embryogenic callus reciprocally, is described. Embryogenic callus could also be produced from anther culture-derived embryoids. Viable protoplasts could be produced easily from embryogenic callus and used in somatic hybridization and direct DNA transfer. Studies on cell selection, cell fusion and genetic transformation in this system are also discussed.
