A SURVEY OF MICROSPORE EMBRYOGENESIS IN LEAF MUSTARD (BRASSICA JUNCEA)

Abstract:
Twenty-four cultivars of leaf mustard (Brassica juncea), representing various morphological and ecological characteristics, were examined for their embryogenic response to microspore culture by applying a method developed in other Brassica species. Induction of microspore embryogenesis was observed in 7 (29.2%) cultivars. Most of responsive cultivars were with early bolting character rather than late bolting character. Thermal shock treatments at 32.5°C for 1 to 3 days prior to incubation at 25°C stimulated embryogenesis. Mean embryo yield was different between cultivars and between donor plant individuals of the same cultivar. The maximum yield of 155 embryos per 100 anthers was obtained in one donor plant of ‘Hsueh Li Hung’, when it was grown under controlled environment from the beginning of bolting and when the thermal treatment for microspores was at 32.5°C for 2 days.
