IN VITRO PROPAGATION OF LUOHANGUO [SIRAITIA GROSVENORII (SWINGLE) C. JEFFERY]

Abstract:
A procedure for the rapid tissue culture of Luohanguo [Siraitia grosvenorii (Swingle) C. Jeffery] is being developed. Nodal explants were induced to grow as multiple-shoot cultures on MS medium containing 1.0mg/l BA, supplemented with 60mg/l adenine and 1.0mg/l IBA. Nodes were removed from the shoots after 4 weeks of growth and subcultured onto fresh culture medium. The effects of plant growth regulators on explant proliferation and development was studied. The results obtained showed that BA greatly stimulated bud formation in concentrations ranging from 0.5–2.0mg/l, but higher concentrations suppressed further bud development. BA was necessary for bud initiation from shoot tip explants. IBA stimulated shoot development in concentrations ranging from 0.5 – 1.5mg/l, but higher concentrations, 2.0mg/l, inhibited bud formation. Adenine greatly stimulated shoot development amd its optimal concentrations would appear to be 40 – 80mg/l.

Under 25°C and higher humidity conditions, the 3 – 4cm long shoots were directly transferred to soil with an approximate 90% survival rate, which could simplify the procedure of in vitro culture, and save expenditure.
