Abstract:
Cherry twisted leaf (ChTL) associated double stranded RNA (dsRNA) was isolated from the petioles of symptomatic Bing cherry plants.
The ChTL-dsRNA was isolated by two cycles of CF11 column chromatography, T1 RNase and DNase treated, and agarose gel purified.
Oligonucleotide primers based on conserved closterovirus sequences were used in RT-PCR to generate a ChTL-specific 600 bp fragment.
This fragment was digoxigenin labelled and used successfully to detect cloned cDNA produced using dsRNA from the putative ChTL-associated virus in Nicotiana occidentalis. Positive results were also obtained when blots containing total RNA from ChTL-affected Bing cherry, and from N. occidentalis infected with the putative ChTL virus were hybridized with the digoxigenin labelled probe.
