Abstract:
Polymerase chain reaction (PCR) technique has been recently applied in Spain to detect phytoplasmas associated to fruit tree diseases, such as apricot chlorotic leaf roll (ACLR), apple proliferation (AP) and pear decline (PD). For PCR reaction a fragment of the 165 rRNA gene was selected as template DNA and specific and non-specific primers were used.
PCR amplification products were analized by restriction fragment length polymorphisms with AluI and RsaI enzymes.
Results provide evidence of the presence of phytoplasmas in fruit trees, as already described for apricot, plum and apple and are the first report of the presence of these pathogens in pear trees in Spain.
