CHARACTERIZATION OF PPV ISOLATES USING MONOCLONAL ANTIBODIES

Abstract:
Monoclonal antibodies, obtained against an Italian apricot isolate of PPV, have been used to characterize a collection of Italian and European isolates, including the two described PPV serotypes: PPV-D from France and PPV-M from Greece.

Monoclonal antibodies produced were against the specific N terminal region of the coat protein, containing the specie-specific epitopes.

Three different serological techniques have been used: DAS-ELISA, Western blot and GIEM. All procedures identified the two PPV serotypes and divided PPV isolates into two groups, corresponding to PPV-D and PPV-M strains, respectively. Moreover, Western blot analysis showed the presence of mixed infections in some isolates.
