Abstract:
As we already presented in the previous two ISHS Fruit Tree Virus Meetings (Thessaloniki, Vienna), we are engaged in pathogen mediated resistance breeding to induce resistance in apricots and plums against plum pox virus (PPV) (Laimer et al., 1989; da Câmara Machado et. al., 1992), PPV certainly being one of the most important pathogens of stone fruit trees in Europe and the whole Mediterranean area.
We have already reported the transformation and regeneration of Prunus armeniaca (Laimer da Câmara Machado et. al., 1992) and P. domestica plants with Agrobacterium tumefaciens strain LBA 4404 containing various binary plasmids, pBinGUSint, carrying the marker gene -glucuronidase (GUS) and pBinPPVm, carrying the coat protein gene of plum pox virus (PPV).
Here we present our efforts to introduce different sequences from the viral genome of PPV into plants to induce resistance.
Also new transformation protocols, which have been developed, allowing an efficient transfer of foreign genes into fruit trees, e.g. different apricot and plum cultivars, will be discussed.
Also our system and some results obtained with in vitro studies on the long term distribution of PPV will be presented.
Encouraged by results of coat protein-mediated protection obtained with herbaceous hosts of PPV (da Câmara Machado et. al., 1992; Regner et al., 1992) we conducted similar challenge infection experiments to investigate resistance phenomena with transgenic Prunus armeniaca and P. domestica. First results of these experiments will be presented.
