DISTRIBUTION OF APPLE CHLOROTIC LEAFSPOT VIRUS IN APPLE SHOOTS CULTIVATED IN VITRO

Abstract:
An immuno tissue-printing protocol has been worked out for the immunological detection of apple chlorotic leaf spot virus (ACLSV) coat protein within in vitro shoots of Malus. Different apple varieties showed a typical distribution pattern for ACLSV in vitro. Accumulation of ACLSV was highest in the basal part of the stem and decreased towards the apex of the shoots. ACLSV reached its maximum amount at the enlarged bases of shoots, where it was unevenly distributed, as tissue was less regularily organized. In cross sections of stem tissue virus coat protein was found within the epidermis, the cortical parenchyma and within the vascular bundles. Within pith tissue virus protein was only detected in adjacent parts of the enlarged bases when shoots were heavily infected. The degree of infection not only influenced the localisation of virus within a cross section, but also its spread into the meristem. Meristems of poorly infected shoots did not contain any visible trays of virus.
