DYNAMICS OF DISSEMINATION OF PRUNUS NECROTIC RINGSPOT VIRUS IN GERMPLASM COLLECTION OF CHERRIES

Abstract:
Cultivars of sweet and sour cherries have been collected in the Research and Breeding Institute of Pomology at Holovousy since 1952. An experimental orchard of sweet and sour cherries was established in 1967 and new cultivars were included into it every year since that time. The assortment was indexed for PNRSV (Prunus necrotic ringspot virus) by ELISA tests in 1983 for the first time. These ELISA tests were made by a technique elaborated by Inotech Diagnostic AG (Swiss) that supplied us with antiserum. Samples of young leaves were collected during the last ten days of May. Altogether 132 cultivars of sweet cherries and 47 cultivars of sour cherries were tested. In 1989 the collection was transferred into a newly established orchard and the same set of cultivars was tested for PNRSV by ELISA tests in 1992–1993 again. Samples of young leaves and blossoms were taken off during May and antiserum obtained from Bioreba (Swiss) was used.

In 1983, 7.9% of sweet cherry trees and 40.7% of sour cherry trees were infected with PNRSV. Regarding cultivars there were 11 sweet cherries and 23 sour cherries infected with the virus. In 1992–1993, 11.0% of sweet cherry trees and 49.5% of sour cherry trees had positive ELISA tests which were from 16 and 45 cultivars respectively. The increase of the natural dissemination of prunus necrotic ringspot virus in the germplasm collections of cherries after 10 years was equal to 3.0% of sweet cherry trees and 8.8% of sour cherry trees. The results indicate also distinctly higher susceptibility of sour cherry cultivars to PNRSV in a comparison with sweet cherry cultivars.
