IN SITU LOCALIZATION OF BLUEBERRY LEAF MOTTLE VIRUS IN BLUEBERRY POLLEN AND ANTHERS BY IMMUNO-GOLD LABELING AND TRANSMISSION ELECTRON MICROSCOPY

Abstract:
In situ localization of blueberry leaf mottle virus (BBLMV) coat protein in blueberry pollen grains and anthers was carried out in ultrathin sections by immunogold labeling and examined in a TEM. When pollen and anthers were obtained from BBLMV-infected blueberry bushes, gold labeling was found in the cytoplasm, intine, and exine of the pollen and in the anther sacs, epidermis and connective tissue of anthers. Statistical comparisons of these samples were made with three different controls: 1. BBLMV- infected tissue labeled with blueberry shoestring virus (BBSSV) antiserum and Protein-A (P-A) gold, 2. BBLMV-infected tissue labeled with preimmune serum and P-A gold, and 3. BBLMV-infected tissue labeled with gold only. When subjected to ANOVA, the number of gold particles found in both pollen and anther samples were significantly different from the controls.
