DEVELOPMENT OF MONOCLONAL ANTIBODIES SPECIFIC FOR STRAWBERRY MILD YELLOW EDGE POTEXVIRUS.

Abstract:
Several strategies were employed for production of monoclonal antibodies (McAbs) specific for strawberry mild yellow edge potexvirus (SMYEPV). Mice were injected interperitoneally with the coat protein of SMYEPV fused to glutathione S-transferase (GEX-CP) or to the IgG binding domain of Protein-A (PRIT-CP). In total four mice were injected with one or both of the fusion proteins. Injection schedules ranged from three to six injections per mouse. Each fusion resulted in thousands of hybridomas but none which produced antibodies specific for SMYEPV were stable. When SMYEPV purified from infected R. rosifolius was used to immunize a mouse, stable hybridomas that produced specific McAbs were obtained. Some of the clones reacted in ELISA with fusion protein as well as virus, some reacted with fusion protein alone and others gave a differential reaction, reacting better with fusion protein than virus. The antibodies have been used to detect the virus in greenhouse and field grown infected strawberry plants by standard triple antibody ELISA.
