DETECTION AND ISOLATION OF STRAWBERRY VEIN BANDING VIRUS IN THE CZECH REPUBLIC

Abstract:
DAS-ELISA with IgG prepared against a Czech isolate of cauliflower mosaic virus (CaMV) was used for pre-screening of strawberries for the presence of strawberry vein banding virus (SVBV) in the Czech Republic. Seventy two samples from 449 tested strawberry plants had extinctions (E405 nm) greater than 0.5. The virus induced symptoms of vein banding and leaf curling after transmission by aphids (Aulacorthum solani Kaltb.) and after leaf grafting onto indicator clones of Fragaria vesca L. Caulimovirus-like particles about 35 nm in diameter were observed in negative stained preparations from leaves of F. vesca clones with SVBV symptoms and E405 nm values in ELISA higher than 0.7. Caulimovirus-like particles about 41 nm were purified from cv. Rujana and Induka. Viral coat protein (48kDa) and DNA (7.9 kbp) were isolated. The virus was identified as the Czech isolate of SVBV.
