Abstract:
Purified nucleic acids from mosaic diseased blueberry tissue contains 2-3 small circular RNAs. cDNA of the major RNA was amplified from nucleic acid preparations by reverse transcription-polymerase chain reaction (RT-PCR) utilizing DNA primers for potato spindle tuber viroid (PSTVd). Cloning and sequencing of the amplified product indicated that the RNA is 351 nucleotides in length and is 41.59% homologous to PSTVd.
This RNA represents blueberry mosaic viroid-like RNA1 (BBMV1). An SP6-generated 32P-labeled cRNA probe of BBMV1 cDNA hybridized to a 351 bp RT-PCR amplified DNA product and to monomeric and multimeric RNA forms of BBMV1 from total nucleic acids of mosaic diseased blueberry tissue.
To our knowledge, this is the first report of isolating and characterization of viroid-like molecules from small fruits.
