Abstract:
A spectrophotometric method for measuring the activity of phenylalanine ammonia-lyase (PAL, E.C. 4.3.1.5) was adapted to apple leaves.
The enzyme is characterized by optima of pH (8.2) and temperature (55°C) and showed its highest activity in the young leaves, levelling down to 20% from the first to the fourth leaf.
Pressure treatment of the leaf discs enhanced the enzyme activity in the light with a peak after 24 hours.
The fact that soluble phenols accumulated later in treated discs than in untreated ones is discussed with reference to metabolic and catabolic processes.
