Abstract:
Paper chromatography in four solvent systems, high sensitivity spray reagents and UV spectroscopy with spectral shifts were used to examine the distribution of major groups of flavonoids in bulked root samples of Lotus pedunculatus 18 days after inoculation (d.a.i.) with Rhizobium loti. Capillary zone electrophoresis (CZE) with spectral array detection was tested for its ability to identify subtle differences in the flavonoid content of micro samples of sterile and bacterially challenged roots.
Bulk tissue analyses indicated major changes in flavonoid biosynthesis in effective root nodules.
CZE analyses provided some inconclusive evidence for bacterially-induced qualitative changes in the flavonoid content of roots during the early phases of infection.
