FURTHER CHARACTERIZATION OF A POTYVIRUS IN ALSTROEMERIA ENDOWMENT SERIES

Abstract:
Alstroemeria Endowment Series exhibited leaf streaking and severe stunting symptoms. Flexuous rods 780 x 12 nm were purified from infected Alstroemeria leaves. Both laminated and pinwheel inclusions were observed in ultrathin sections from infected Alstroemeria leaves. With indirect ELISA, the virus did not react to alstroemeria mosaic virus antiserum and a broad spectrum potyvirus monoclonal antibody. However, the dissociated viral coat protein reacted strongly to a potyvirus group specific polyclonal antibody in western immunoblot. Since attempts to transmit this virus mechanically from Alstroemeria to other common virus indexing plants failed, we were unable to obtain single lesion isolates. Based on the differences in coat protein molecular weights from two batches of viruses purified from infected Alstroemeria leaves, the possibility of a mixed virus infection involved in this Alstroemeria leaf streaking disease thus cannot be ruled out.
