TOWARDS A RELIABLE AND FAST IMMUNOLOGICAL DETECTION OF TOMATO SPOTTED WILT VIRUS IN INFECTED TISSUES

Abstract:
Different enzyme-immunoassays were evaluated for the detection of tomato spotted wilt virus (TSWV) in plant extracts. Immunoelectron microscopy was used to substantiate the results of the other immunological methods. Biotin mediated enzyme-linked immunosorbent assay (ELISA) procedures were more sensitive than the standard ELISA procedure which uses virus specific antibodies labelled with alkaline phosphatase. Goat antibodies to biotin labelled with alkaline phosphatase (GAB-AP) proved to be a useful immunological probe to detect the bound biotinylated TSWV-specific polyclonal antibodies in ELISA, dot immunobinding assays, and tissue-print immunoassays. The later method has the advantage of enabling the specific diagnostic with the localisation of the virus in the tissues. In addition, tissue-print immunoassay is a simple, rapid and cheap method, suitable for testing both small and large numbers of samples. Therefore, it can prove to be a powerful tool for critical diagnosis in quarantine cases, and for plant health certification programmes.
