| Abstract:
A permanent screenhouse was constructed for the new system of plum maintainance breeding in RIFG Holovousy. The propagation stocks for the production of buds and grafts were founded. An experimental orchard for clone verification was planted. The healthy plum material was prepared for planting of a seed orchard on a limited area. Basic virus-free material of plum cultivars and rootstocks is output of the research programme: The research of plum virus elimination methods. In vivo and in vitro methods were used for elimination PPV, PNRSV and PDV from plum clones. Four combine elimination techniques displaied different efficiency. Two collections of virus-free material of plum varieties and rootstocks are maintained in the permanent screenhouse. The first collection of virus free clones was obtained by ELISA indexing of PPV, PNRSV, PDV and ApMV from plums main tained in the natural isolation plot Jelec. The second collection was obtained by in vivo and in vitro combined techniques often connected with in vivo and in vitro thermotherapy and chemotherapy. The efficiency was checked by ELISA methode. | |
