ISHS Acta Horticulturae 356: II International Symposium on Olive Growing

ATTEMPTS TO GENERATE HAPLOID PLANTS FROM IN VITRO CULTURES OF OLEA EUROPAEA L. ANTHERS.

Authors:   E. Perri, M.V. Parlati, R. Mule, A.S. Fodale
Abstract:
This study is a further contribution to obtaining haploid olive plants from anthers. Media with different amounts of growth regulators and two culture conditions were used. In medium P, based on MS salts plus 5 mg/l of thidiazuron (TDZ) and 0.1 mg/l of 1-naphtaleneacetic acid (NAA), in the dark at 27°C, abundant callus proliferation was obtained in 12% of the explants, better than medium I (Mulè et al., 1992) and better what has been obtained up to now in this research program. Stabilization of the culture and further callus development is still problematic.
