PROCEDURE TO STUDY OVULE SENESCENCE IN OLIVE

Abstract:
Aniline blue fluorescence method was found to be an useful tool to discriminate viable from senescent ovules and to determine the timing and factors influencing ovule senescence in olive. Olive viable ovules stained with aniline blue exhibited an orange-colour with a light yellowish fluorescence limited to micropylar area. In constrast, senescent ovules showed an intense yellow fluorescence over their entire surface. Following to fertilization, the seed growth accelerated the degeneration process in the unfertilized ovules of the same ovary, but also in adjacent flowers. Differences in longevity among ovules of the same flowers were smaller than among ovules of different flowers. However, it is not unusual to find viable and senescent ovules within the same ovary. This whole of observations have been summarized in order to propose a procedure to evaluate ovule viability in olive.
