ISHS Acta Horticulturae 356: II International Symposium on Olive Growing

PRELIMINARY STUDIES ON ISOENZYMATIC AND ENZYMATIC ACTIVITIES ISOLATED FROM OLIVE PLANTS.

Authors:   M. Patumi, G. Fontanazza, R. Nucci, C. Vaccaro
Abstract:
For isoenzymatic and enzymatic activity studies it is very important to obtain a proteic extract whose concentration and stability, during tissues conservation and extraction processes can be certainly assessed. This work reports some considerations arised by the sperimentation carried out in our Istitutes.

In the attempt to get fastly in solution proteins from different tissues (leaves, roots and fruits) we have used soaking, grinding and sonication techniques without reproducible results probably because of interfering substances or/and of modified proteins.

No result we got in presence of reducing agents and of the insoluble polyvinylpirrolidone for adsorbing phenols.

For the above reasons we are in progress an extraction from cellular organelles obtained by cellulolitic enzymes and osmotic shock that will allow the isolation of the enzymatic activities under study.
