ISOLATION AND CULTURE OF BLACKBERRY PROTOPLASTS (Rubus sp.) VAR. "HULL THORNLESS".

Abstract:
Blackberry var. "Hull Thornless" protoplasts were isolated from leaves and petioles of proliferating in vitro cultures. Protoplasts were cultured on K8P medium supplemented with 2,4-D, NAA, BA and TDZ as liquid or agarized bead type cultures. Different responses were obtained in terms of the efficiency for first division, colony and microcallus formation. After 60 days in the bead type culture it was possible to produce large quantities of callus, that thereafter proliferated on an MS-based medium. On the contrary only few colonies were obtained in liquid medium. Different regeneration media based on MS medium supplemented with 2,4-D, BSAA, BA, and enzymatically hydrolisated agar were tested, but no organized structures were found after several months in culture.
