ORGANOGENESIS AND SOMATIC EMBRYOGENESIS FROM IMMATURE EMBRYOS OF BLACKBERRY

Abstract:
Immature embryos with part of the endosperm, derived from self-pollinated seeds of 'Hull Thornless', 'Thornfree' and 'Comanche', were aseptically excised about 28 days after pollination. The material was placed for 5 min in a solution of 1% citric acid to avoid oxidation and then transferred on MS salts and vitamines medium with 3% sucrose, 0.7% agar Bacto-Difco, different combinations of various growth regulators (IAA, NAA, 2,4-D, 2iP, BA, Zea, GA3) and with or without 400 mgl-1 casein hydrolysate. In 'Hull Thornless' part of the material was pretreated for 10 min in a solution containing 4 mgl-1 2,4-D before placing on solidified media with only BA.

Different morphogenic processes were observed in relation to the genotype and the culture medium. Shoots were obtained through direct organogenesis from cotyledons and through organogenesis after callus induction; plantlets were obtained after somatic embryogenesis in 'Thornfree'. The plantlets from the same seed showed an evident phenotypic variation. In 'Hull Thornless' 2,4-D pretreatment highly promoted differentiation.
