ISHS Acta Horticulturae 341: IV International Mango Symposium

EFFECT OF GROWTH REGULATOR AND MEDIA ON IN VITRO SHOOT TIP CULTURE OF DIFFERENT CULTIVARS OF MANGO (MANGIFERA INDICA L.) ROOTSTOCKS

Authors:   Z. Yang, P. Lüdders
Keywords:   in vitro, Mangifera indica, shoot tip, growth regulator
Abstract:
Growth regulators, media components, cultivars that affect shoot tip establishment and growth of mango-rootstock (Mangifera indica L.) in vitro were studied. These included the role of growth regulators, basal medium formulations, and genotypes. Shoot growth was better in G medium containing a combination of BA 1.0 mg/l, zeatin 1.0 mg/l, 2iP 2.0 mg/l, IAA 1.0 mg/l and IBA 0.5 mg/l plus CH 300 mg/l, glutamine 300 mg/l and 3 % sucrose than either B-5 basal medium or woody plant medium (WPM). The addition of GA3 (5–10 mg/l) to the medium did not improve shoot growth. 'Terpentine' gave the best results in shoot tip establishment and elongation compared to other cultivars tested. The highest percentage of proliferated shoots was obtained from explants in May and June in comparison with explants from other months. Chemical names used: N-(phenylmethyl)-1H-purine-6-amine (BA); (2-methyl-4-(1H-purine-6-ylamino)-2-buten-l-ol (zeatin, ZT); 6-furfurylaminopurine (kinetin, KT); N-(3-methyl-2-butenyl) -1H-purine-6 amine (2iP); l-H-indole-3-acetic acid (IAA); 1H-indole-3-butyric acid (IBA); 1-naphthaleneacetic acid (NAA); gibberellic acid (GA3); casein hydrolysate (CH)
