ISOZYMIC ANALYSIS OF SOMACLONAL VARIATION AMONG REGENERANTS FROM APPLE ROOTSTOCK LEAF TISSUE

Abstract:
Isozymic analysis was used to investigate genetic variation of 5 apple clonal rootstocks (M.9, M.26, MM.106, MM.111 and Mark). Analyses were performed using leaf tissue collected from plants actively growing in vivo and in vitro. Isozyme polymorphism of several in vivo growing regenerants from leaf callus of M.26 and MM.106 was also evaluated. Nine enzyme staining systems for each clone: PGN, PGI, EST, APH, GDH, ENP, LAP, MDH and GOT were considered. Wide polymorphic variability was observed with APH and PGI systems and a clear discrimination between the five rootstocks waspossible. Some patterns seemed to be influenced by the growing conditions of the plant material: in vivo versus in vitro. Isozyme polymorphism was observed among regenerants and based on banding patterns, rootstocks and regenrants could be distinguished.
