TRANSFORMATION OF POTATO (SOLANUM TUBEROSUM) WITH A GENE FOR AN ANTI-BACTERIAL PROTEIN, CECROPIN

Abstract:
Leaf discs and microtuber slices of two potato cultivars were co-cultivated with Agrobacterium tumefaciens LBA4404 containing an antibacterial gene, cecropin, fused to a plant secretory signal sequence and cloned into the binary vector pB1121. Four regenerants of 'Atlantic' and eight of 'FL1607' exhibited kanamycin resistance. Transformation of these 12 lines was confirmed by histochemical assays for beta-glucuronidase activity in leaves and tubers. Bacterial challenge experiments with potato tuber soft rot pathogens, Erwinia carotovora subsp. carotovora and E. carotovora subsp. atroseptica, revealed a moderate (10–20 %) decrease in maceration among the transformed lines, compared to cultivar controls. However, because only a limited number of greenhouse-grown tubers were tested, further testing, including tests of field- grown tubers, is necessary to identify individual transformed lines with resistance.
