PRODUCTION OF MEDICINAL ALKALOIDS BY PAPAVER BRACTEATUM CULTURED CELLS

Abstract:
The Papaver species have become one of the most interesting subjects in the field of plant cell culture for problems of secondary metabolism. In 1979 Kuzovkina and Rabinovich have obtained Papaver bracteatum tissue culture. The strain, characterized by stable growth, was selected for our experiments. This strain has been shown to be able to produce only benzophenanthridine alkaloids. To establish new cell lines both the optimization of nutrient media and different concentrations and combinations of phytohormones were used. As a result of prolonged selection several callus culture variants were obtained. Two of them possess the ability to grow on the nutrient medium lacking hormones. These cultures show organogenic differentiation growth: root formation, initiation of shoot buds and formation of great number of adventitious embryos. Spectrophotometric and TLC analysis revealed differences in alkaloid spectra in all strains studied. The production of protopine, benzophenanthridine and morphinan alkaloids in obtained cell lines is demonstrated.
