AGROBACTERIUM - MEDIATED TRANSFORMATION OF LILIUM

Abstract:
Monocotyledons are generally considered to be insensitive to Agrobacterium infection and subsequent transformation. When bulb-scale sections of Lilium longiflorum cv. 'Nellie White' were co-cultivated with a tumor-inducing strain of Agrobacteriun tumefaciens-C58, small tumor-like swellings developed. Analysis of callus homogenates revealed the expression of the T-DNA encoded genes for nopaline indicating transformation of the Lilium tissue. Untreated Lilium scales and those cocultivated with disarmed strains of A. tumefaciens developed little callus and showed no evidence for opine synthesis. Cocultivation with three different strains of A. rhizogenes did not induce the formation of the typical hairy-root, but rather generated the development of an abundance of tumor-like callus. No opine synthesis was evident in any of the A. rhizogenes-induced tumors. The results indicate that transformation of Lilium depends on the Agrobacterium strain used and clears the way for the future production of transgenic lilies using engineered strains of C58 or similar biovars.
