CONTROL OF MORPHOGENESIS IN THIN CELL LAYER EXPLANTS OF MUSCARI BOTRYOIDES MILL.

Abstract:
The effect of different concentrations of the IBA and BA on the control of morphogenetic processes in small epidermal and subepidermal explants of Muscari botryoides Mill. were investigated. Depending on the season, the regeneration ability of explants on basal media amounted from 10% (September) to 56% (May) and each explant formed on average 1–2 bulblets. Further increase in both regeneration frequency and number of bulblets formed required the presence of both classes of growth regulators, i.e. auxin (0.2–2.0 mg/dm3 IBA) and cytokinin (1.0–2.0 mg/dm3 BA). The diameters of regenerating bulblets depended on auxin concentration in the medium. An increase in IBA concentration resulted in proportional increase in the bulblet diameter as well as their fresh and dry weight. Auxin stimulated development and elongation of leaves of newly formed bulblets, it also promoted root and callus formation, but only slightly affected a bulblet differentiation. Benzyladenine applied together with auxin induced bulblet differentiation, it inhibited, however, growth processes and root formation. The dose of 2.0 mg/dm3 IBA neutralized growth retardation. provoked by cytokinin and caused an increase in bulblet diameters promoting simultaneously the bulblet differentiation.
