A METHOD FOR DISINFECTION OF UNDERGROUND RHIZOME TIPS OF ALSTROEMERIA AND HELICONIA.

Abstract:
Disinfection of initial explants is a major problem when micropropagating species where underground rhizomes are the only source of axillary buds. A procedure for disinfection of rhizome tips has been developed, based on trimming of scale leaves 3 times alternating with short (1 – 10 min) immersions in disinfectants. Yields of growing, axenic explants of 80 and 60 % were obtained in Alstroemeria and Heliconia, respectively when using all 3 steps. When omitting one step significantly higher rates of contamination were observed in Alstroemeria. Rhizome tips shorter than 4 cm, from fast growing stock plants, gave the lowest contamination in Heliconia.
